Melanocortin Receptors: A Short Research Primer

What the five melanocortin receptor subtypes are, which peptides are used as reference agonists in the literature, and how selectivity is characterized in cell-based assays.

Five receptors, one peptide family

The melanocortin system consists of five G protein-coupled receptors, MC1R through MC5R, that respond to peptides derived from proopiomelanocortin. Because the natural ligands overlap across subtypes, most of the pharmacology literature is concerned with a single question: how selectively does a given analog engage one receptor over the others?

In laboratory work this is answered with binding assays and functional readouts — typically cyclic AMP accumulation in cells engineered to express one receptor subtype at a time. A compound is described as non-selective when it produces comparable activity across several subtypes, and selective when its potency differs meaningfully between them.

Reference compounds used in receptor panels

Cyclic α-MSH analogs appear throughout this literature as reference agonists precisely because they are non-selective: their activity across subtypes gives a stable baseline against which newer, more selective analogs are compared. Melanotan II is the analog most often cited in that role.

Compounds characterized as MC3R/MC4R-preferring, such as bremelanotide (PT-141), are used in the opposite way — to isolate signaling attributable to a narrower subset of receptors. Pairing a non-selective and a subtype-preferring agonist in the same experiment is a common design for separating subtype contributions.

What these studies do and do not establish

Receptor-level findings describe molecular interactions in a controlled system. They do not establish physiological, cosmetic, or clinical outcomes, and results in cell culture frequently do not transfer to whole-organism models. Laboratory documentation should keep those layers separate.

For research handling, the practical variables that most affect reproducibility are peptide purity and identity for the lot in question, reconstitution accuracy, and storage discipline — not the choice of analog.
