Reconstitution and Storage of Lyophilized Research Peptides

Diluent selection, concentration math, labeling discipline, and the storage variables that determine whether a lot behaves consistently across experiments.

Diluent and technique

Lyophilized peptides are reconstituted with a compatible sterile diluent — commonly bacteriostatic water, which contains 0.9% benzyl alcohol and permits multiple withdrawals from one vial. Direct the stream against the glass wall rather than onto the cake, and allow the material to dissolve without shaking; mechanical agitation can denature sensitive sequences and introduce foaming that makes accurate withdrawal difficult.

If the solution does not clear within a few minutes, warm it gently to room temperature and swirl. Persistent cloudiness or visible particulate after full dissolution is a flag to check the lot documentation rather than to proceed.

Concentration math and labeling

Concentration is vial mass divided by diluent volume. 10MG in 2 mL is 5 mg/mL; the same 10MG in 1 mL is 10 mg/mL. Write the resulting concentration, the diluent volume, and the date directly on the vial at the moment of reconstitution — reconstructing it later from memory is where most traceability failures begin.

For blended vials, label each component's concentration separately rather than the total mass, so downstream calculations cannot silently use the wrong figure.

Storage and stability

Sealed lyophilized vials are the most stable form: kept refrigerated, dry, and protected from light, they tolerate long storage well. Once in solution, peptides are considerably less stable and should be kept cold and used within the window your protocol defines.

Repeated freeze–thaw cycling is the most avoidable cause of degradation in reconstituted material. Aliquot into single-use volumes before freezing so no vial is thawed more than once, and keep a written log tying each aliquot back to its lot number and certificate of analysis.
